C3 Pathway and the First Stable Product: 3-PGA (Calvin Cycle)

Biology · Photosynthesis in Higher Plants · NEET

The C3 pathway (Calvin cycle) fixes CO2 onto a 5-carbon acceptor called RuBP using the enzyme RuBisCO. This makes two molecules of a 3-carbon acid, 3-phosphoglyceric acid (3-PGA), which is the first stable product. Memory hook: "C-THREE = first product has THREE carbons (3-PGA)." So the cycle is named C3 because its very first stable product is a 3-carbon compound.
C3 Pathway: Carboxylation StepRuBPacceptor (5C)RuBisCOcarboxylase2 x 3-PGAFIRST STABLEPRODUCT (3C each)+ CO2RuBP (5C) + CO2 (1C) -> two 3-PGA (3C + 3C) = 6 carbons balance
Carboxylation in the C3 pathway: RuBisCO adds CO2 to the 5-carbon acceptor RuBP, forming two molecules of the 3-carbon first stable product, 3-PGA. The carbons balance: 5 + 1 = 3 + 3.

Your doubts, answered

Is 3-PGA the first stable PRODUCT or the first CO2 ACCEPTOR?

They are different molecules and NEET loves to swap them. The first CO2 ACCEPTOR is RuBP (ribulose-1,5-bisphosphate), a 5-carbon sugar. The first stable PRODUCT is 3-PGA (3-phosphoglyceric acid), a 3-carbon acid. RuBP is the input that grabs CO2; 3-PGA is the output made just after CO2 is fixed.

Why is the Calvin cycle called the C3 pathway?

Because the first stable product formed after CO2 fixation is a 3-carbon molecule, 3-PGA. The pathway is named after the number of carbons in its first stable product, not after the acceptor (which has 5 carbons). Plants that use only this pathway are called C3 plants.

Which enzyme makes 3-PGA and how many form per CO2?

The enzyme is RuBisCO (RuBP carboxylase-oxygenase). It adds one CO2 to one RuBP. For every single CO2 fixed, TWO molecules of 3-PGA are formed. So the carbon count balances: RuBP (5C) + CO2 (1C) = 6 carbons = two 3-PGA (3C each).

Is RuBP the same as 3-PGA?

No. RuBP is a 5-carbon sugar phosphate that acts as the acceptor. 3-PGA is a 3-carbon acid that is the first stable product. Do not confuse RuBP (ribulose bisphosphate) with 3-PGA (phosphoglyceric acid) even though both appear in the carboxylation step.

OAA is also a first product in photosynthesis. When is it PGA and when is it OAA?

In the C3 (Calvin) pathway the first stable product is the 3-carbon 3-PGA. In the C4 (Hatch and Slack) pathway the first stable product is the 4-carbon OAA (oxaloacetic acid). If the plant is C4 (like sorghum, maize, sugarcane), the first product is OAA; if C3, it is PGA.

⚠️ The NEET trap
3-PGA is the first CO2 acceptor of the Calvin cycle.
3-PGA is the first stable PRODUCT; the first CO2 ACCEPTOR is the 5-carbon RuBP.
🧠 Acceptor RuBP has 5 carbons; product PGA has 3 carbons. Product decides the name C3, not the acceptor.

Real NEET questions

NEET 2021

The first stable product of CO2 fixation in sorghum is:

A · Succinic acid
B · Phosphoglyceric acid
C · Pyruvic acid
D · Oxaloacetic acid
Solution: This tests whether you know PGA is the C3 first product but the plant here is C4. Sorghum is a C4 plant, so its first stable fixation product is the 4-carbon oxaloacetic acid (OAA), not the 3-carbon PGA. Phosphoglyceric acid (PGA) would be the first product only in C3/Calvin plants. Answer: OAA (D).
NEET 2026

The enzyme required for carboxylation in the Calvin cycle is

A · Hexokinase
B · PEP carboxylase
C · RuBP carboxylase - oxygenase
D · Carboxypeptidase
Solution: Carboxylation in the C3/Calvin cycle is catalysed by RuBP carboxylase-oxygenase (RuBisCO), which fixes CO2 onto RuBP to form two molecules of 3-PGA. PEP carboxylase is the C4 mesophyll enzyme (a distractor). Answer: RuBP carboxylase-oxygenase (C).
NEET 2025

Which of the following statements about RuBisCO is true?

A · It is an enzyme involved in the photolysis of water.
B · It catalyzes the carboxylation of RuBP.
C · It is active only in the dark.
D · It has higher affinity for oxygen than carbon dioxide.
Solution: RuBisCO catalyses carboxylation: CO2 is added to the 5-carbon RuBP to form two molecules of 3-PGA (the first stable product of the C3 pathway). Water photolysis is done by PS II, and RuBisCO actually has a higher affinity for CO2 than O2. Answer: It catalyzes the carboxylation of RuBP (B).

Frequently asked

What is the first stable product of the C3 pathway?

3-phosphoglyceric acid (3-PGA), a 3-carbon acid. Two molecules of it are formed for every CO2 fixed onto RuBP.

Why is it called the C3 pathway?

Because the first stable product formed after CO2 fixation is a 3-carbon compound, 3-PGA. The pathway and the plants that use it are named after this 3-carbon product.

What is the CO2 acceptor in the C3 pathway?

The primary CO2 acceptor is RuBP (ribulose-1,5-bisphosphate), a 5-carbon molecule. It is not 3-PGA; 3-PGA is the product, not the acceptor.

Who discovered the C3 pathway?

Melvin Calvin, which is why the C3 pathway is also called the Calvin cycle.

How is 3-PGA different from OAA?

3-PGA (3 carbons) is the first stable product in C3 plants; OAA (4 carbons) is the first stable product in C4 plants. Number of carbons and pathway are the key difference for NEET.
